Linee di ricerca

The research topics are mainly focused on neoplastic pathology (breast, head-neck, neuro-endocrine system, central nervous system ) and on muscle pathology. The multicentricity and multifocality of breast ductal carcinoma is investigated through the study of clonality with mitochondrial DNA analysis and through the concept of field cancerization especially on resection margins. Moreover, the diagnostic and prognostic values of DNA methylation analyzing several target genes by quantitative bisulfite next-generation sequencing and miRNAs analysis in oral carcinoma (follow-up of pre-neoplastic lesions using genetic markers on scraping to identify patients to be biopsied) is evaluated. Finally we investigate potential prognostic and predictive value of morphological / biomolecular markers to predict survival and response to therapy in high grade osteosarcoma of maxillary bones. Central Nervous System: one focus of research is on the evaluation of the prognostic impact of Galectin 3 on ACTH pituitary . Another is the correlations between morphology of recurrences of glioblastoma multiforme (GBM) and response to radio-chemotherapeutic treatment. The research in muscle pathology mainly addresses to the study of etio-pathogenetic mechanism in muscle dystrophies with specific interest in muscle dystrophy 1F, a rare disease with dominant autosomic inheritance by ultrastructural study on patient biopsies and an in vitro study by WB, CRISPR-CAS 9 and immunohistochemistry (IF at confocal microscopy). Mutation on Transportin 3 gene has been recently identified (Transportin 3 protein of the importin/exportin family) . A relationship between nucleus and cytoskeletal system has been established with likely important role in functional alteration allowing to muscle atrophy. On the same in vitro model in collaboration with prof Angelini’s group (IRCCS S.Camillo, Ve) we are studying the expression of the muscle-specific miRNA during myoblast differentiation related to Transportin 3 expression. Transmission electron microscopy is constantly a useful tool in both SNC and muscle pathology studies
