LINEE DI RICERCA

Study of the role of mitochondria and the mitochondrial genetic system in the aging process and in age-related pathological conditions (diabetes, neurodegenerative diseases, cancer etc…) as well as of the nutritional strategies or drugs that may prevent the degenerative processes, eventually promoting mitochondrial biogenesis. Determination and quantification of mtDNA damages as well as analysis of mtDNA stability and TFAM-mtDNA interactions. Study of nuclear factors involved in the signaling pathways of mitochondrial biogenesis (NRF-1, PGC1). Identification of nuclear-mitochondrial genomic profiles involved in the pathogenesis of common age-related diseases, diagnostic and therapeutic implications through pharmacological prevention. The experimental models are: tissue samples from rats and humans of different ages, rats treated with calorie restriction diet (collaboration with Prof. C. Leeuwenburgh, University of Florida, Gainesville, FL, USA) either compounds qualified as nutraceuticals or similar as well as cancer tissues samples

Study of the human mitochondrial variability in the mitochondrion and in the nucleus. Such  research involves the usage of the data bank HmtDB (www.hmtdb.uniba.it), the production of the compilations of nuclear sequences of human mitochondrial origin (Human NumtS) and also of other organisms, the development of bioinformatics systems for the prediction of pathogenic mitochondrial mutations and for the prediction of human mitochondrial haplogroups.

Studies on Pathogenetic Mechanisms responsible for the Leber Hereditary Optic Neuropathy. Role of mitochondrial biogenesis in modulating the disease penetrance and as predictive tool for the clinical manifestation of the disease.

Investigations on nucleus-mitochondrial interaction in health and in disease as well as on the role of members of mTERF family protein in the expression of mitochondrial genes by using Drosophila melanogaster and sea urchin as model system
