LINEE DI RICERCA

Combined use of high resolution multinuclear multidimensional NMR, circular dichroism, fluorescence spectroscopy and molecular dynamics calculations to study the structure and dynamics of proteins and peptides. The research aims to determine the 3D structure and the molecular dynamics of proteins and peptides in order to correlate these features with their biological functions. The techniques used for structure determination are mainly multinuclear and multidimensional high resolution NMR, circular dichorism and fluorescence spectroscopy. As for the study of their dynamics we use NMR, molecular dynamics calculations and fluorescence spectroscopy. The systems we are interested in are: 1) Proteins belonging to the lipocalin family such as Major Urinary proteins, a pheromonal protein and a potent human allergen. Main goal is the production of a vaccine. 3) A protein belonging to the Fatty acid binding proteins family from the parasite S. Mansoni. The study aims to develop a vaccine against schistisomyasis. Main goal is the production of a vaccine. 3) Proteins produced by pathogenic fungi that exert a toxic effect on plant such as platanus or tomatoes. The objective is to understand its mechanism of action. 4) Proteins of unknown functions derived from the genome of the plant pathogen X. citri that affect citrus trees. Objective of this study is to understand their role in the pathogen physiology. 5) Toxins derived from the venom of scorpions and active on voltage-dependent K+ channels. Main goal is to produce molecules for therapeutic use against autoimmune diseases and allograft rejection. 6) Antimicrobial peptides from mammal, amphibian or insect origins. To understand their mechanism of action and to develop new and more active drugs. 7) We use some of these proteins, especially the lipocalins and the fatty acid binding proteins, as models for protein unfolding/refolding studies. Most of the proteins we use are recombinant proteins that we clone, express and purify in the laboratory. Most of the peptides, instead, are synthetic and are produced by two groups in Brazil we are collaborating with.
