1. Structure and Dynamics of Biopolymers by NMR spectroscopy and computational chemistry. Research aims: development of spectroscopic and computational methodologies for the study of complex biological systems. Our group deals with the quantitative measurement of geometrical (torsion angles and interatomic distances) and dynamical parameters (overall and internal correlation times), to be utilised as restraints in molecular dynamics simulations of solvated systems. The three-dimensional structure and the intrinsic mobility of macromolecules in solution are actually two important features for a deep understanding of molecular recognition mechanisms. 2. Studies on DNA-xenobiotic Molecular Recognition for the design of new antitumour agents. Research topics: QSAR studies on novel topoisomerase inhibitors. Photodynamic activity of perylenequinones. Studies on the interaction between oligonucleotide and N-heterocyclic systems. Structural analysis of ligand-DNA complexes, together with association constants and mean residence time measurements. 3. Conformational Analysis of natural and synthetic peptides with potential antitumoural activity. Research topics: Structure-function relationship studies on serine protease inhibitors from leguminous plants. Anti-angiogenic peptides. 4. Isolation and Structural Characterisation of biologically active secondary metabolites. Research aims: novel natural substances endowed with antifeedant and insecticide activity extracted from forage plants. Many plants produce a wide variety of secondary metabolites with toxic, repellent or antifeedant activity towards phytophagous insects, which may be utilised with success in agriculture. Our research aims at the screening of selected secondary metabolites (“metabolic profiling”) by their structural elucidation and measurement of their efficacy againts agriculturally relevant insect species.
